An important role in skin hydration is played by hyaluronic acid (HA), which is one of the main components of the extracellular matrix of the skin. Efficacy and safety studies of unmodified HA at various concentrations
In order to study the effectiveness and safety of the use of unmodified HA of various concentrations, when conducting intradermal injections, the scientific supervisor of OKK-EXPERT Ph.D. Larkina Svetlana with co-authors conducted morphological studies of the skin of experimental animals (rabbits). The studies were carried out on the basis of the histological laboratory of the Department of Pathological Anatomy with a sectional course of the Odessa National Medical University. For the study, 4 biorevitalizants with different concentrations of HA containing mannitol were selected: Stylage Hydro and Luminera Hydryal 2%, 3%, 4%.
After the administration of biorevitalizants with mannitol containing various concentrations of HA, the following results were observed (skin samples were taken after 2 weeks, 1 and 2 months after injections):
- Morphological changes were observed in all samples.
- No degenerative or atrophic processes were found.
- There was an increase in the number of connective tissue fibers in all layers of the dermis and in all samples, compared with the control preparation.
- When using 1.4% and 2% HA solution, more pronounced changes were observed in the papillary layer. When using a 3% and 4% solution of hyaluronic acid — changes were expressed in all layers of the dermis, but to a greater extent in the reticular layer.
- 2 weeks after administration of 4% HA solution: an increase in the number of fibroblasts in the papillary dermis was noted, almost 4 times compared with the control sample.
- Drugs with a lower concentration of HA (1.4% and 2%) gave more pronounced results in the early stages (2 - 4 weeks), and drugs with a high concentration of HA (3.0% and 4.0%) — at a later date (8 weeks).

The conducted studies indicate that the formation of connective tissue fibers of the dermis increased with an increase in the concentration of hyaluronic acid, which may indicate a greater compaction of the dermis when using highly concentrated biorevitalizants. It is also possible to assume the relationship between the concentration of HA and morphological changes in the skin: the higher the concentration, the later the biorevitalizing effect occurs, which may indicate that the stimulating effect of HA occurs when its physiological concentration in tissues is reached. The antioxidant activity of mannitol provides a slowdown in the biodegradation of HA, which may serve as a basis for reducing the number of necessary procedures.
